NMN supplement: what the human evidence actually shows

Quick answerNMN (nicotinamide mononucleotide) is a precursor of NAD+ and is sold as an anti-aging supplement. Human trials are short and small. A 2026 meta-analysis of 15 trials found NMN at 250 to 2,000 mg a day was well tolerated for up to 24 weeks, but showed no clear benefits for weight, blood sugar or cholesterol, only a slight drop in diastolic blood pressure. A 2025 meta-analysis found no benefit for muscle mass or strength in older adults. Claims of reversing aging are not supported.

What NMN is

Nicotinamide mononucleotide is a precursor of nicotinamide adenine dinucleotide (NAD+). A 2025 review in the Journal of Cachexia, Sarcopenia and Muscle notes that NMN and the related compound nicotinamide riboside (NR) have drawn interest for raising NAD+ and possibly slowing age-related muscle loss in preliminary models.

NMN is sold as capsules, powders and gels, often labeled as beta-nicotinamide mononucleotide.

What human trials show

OutcomeMeta-analysis result
Safety (10 of 15 trials)No increase in adverse events or liver enzymes
Weight, BMINo significant effect
Fasting glucose, HbA1c, cholesterolNo significant effect
Diastolic blood pressureSlight decrease
Muscle mass, grip, gait speedNo significant effect in older adults

Metabolism and safety. A 2026 meta-analysis in Nutrients included 15 randomized trials, using 250 to 2,000 mg a day for 14 days to 24 weeks. NMN did not raise adverse events or the liver enzymes ALT and AST. Broad metabolic benefits were not evident, though diastolic blood pressure fell slightly and insulin resistance showed a non-significant downward trend.

Blood sugar and lipids. A 2024 meta-analysis in Current Diabetes Reports reviewed 8 trials with 342 mostly healthy middle-aged and older adults and found no significant benefit on fasting glucose, insulin, HbA1c, insulin resistance or lipids.

Muscle. The 2025 muscle meta-analysis, in people with an average age over 60, found NMN did not improve muscle index, grip strength, gait speed or chair stand time. The authors concluded current evidence does not support NMN or NR for preserving muscle in older adults.

Questions people ask

What is NMN supplement good for?
It is an NAD+ precursor, but human meta-analyses have not shown clear benefits for weight, blood sugar, cholesterol or muscle.
Is NMN safe?
Short-term trials up to 24 weeks found it well tolerated with no rise in liver enzymes. Long-term safety is unknown.
How much NMN should I take?
Follow the label. Trials used 250 mg to 2,000 mg a day; NOW Foods' capsule is 250 mg, 1 to 2 times daily.
Does NMN reverse aging?
No human evidence shows that. Muscle and metabolic outcomes did not improve in meta-analyses.
NMN or NR: which is better?
Both are NAD+ precursors. Neither preserved muscle in older adults in a 2025 meta-analysis.
